1.0 Introduction

The addition of the Lambda Plate Compensator Kit to your Leica CM E or DM E microscope provides

additional capabilities such as enabling quick yet accurate differential of gout/pseudo gout.

NOTE: It is not possible to use the DM E with Kohler Illumination and the Lambda

Plate Compensator.

2.0 Installation

Installation of the kit is a simple, straight forward operation. Care should be taken, however, to

ensure proper alignment of the parts relative to one another and the microscope stage.

2.1 Analyzer

1. Remove the microscope viewing body by loosening the locking screw.

2. Insert the analyzer into the recessed area of the stand.

3. The locating pin on the analyzer should mount into the center opening in the stand cutout.

4. Mount the viewing body on the stand with the eyetubes facing the front of the microscope.

Tighten the locking screw.

NOTE: The analyzer should be placed between the microscope stand and any additional

accessories such as multiple viewing adapters, aperture viewers, or filters. The tube

would then be mounted above the accessory in use.

2.2 Lambda Plate Compensator

1. Raise the condenser to it’s upper stop position.

2. Place the compensator onto the illuminator window assembly (Figure 1).

3. Orient the polarizer assembly approximately (Figure 2).

2.3 Orienting the Lambda Plate Compensator

1. Rotate the Lambda Plate Compensator out of light path (dotted line in Figure 2).

2. Turn on the illuminator.

3. Field should be nearly (or totally) black, this is called extinction.

4. Rotate the polarizer until maximum extinction (blackest field) is achieved. This should require

only slight movement in one direction or another. If not, double check original mounting

position.

5. Tighten the locking screw.

3.0 Operation

3.1 Brightfield

For brightfield viewing remove the analyzer from the stand by loosening out the locking screw

and taking off the vieiwng body. Remove the Lambda Plate Compensator from the illuminator

window by loosening the locking screw.

3.2 Normal Polarization

Once the polarizer is correctly orientated (Section 2.3), the microscope is set up for polarized

light. Swing the full wave plate out of the light path.

4.0 Differentiation of

Gout/Pseudo Gout

The following section explains the basic procedure for gout/pseudo gout differentiation. This test

is made possible due to the negative birefringence of urates and positive birefringence of

pyrophosphates. Both gout (monosodium urate) and pseudo gout (calcium pyrophosphate)

crystals tend to be needle shaped. However, many crystals may be broken and/or irregular. To do

the test, it is necessary to find at least one intact crystal orientated North-South (i.e., vertically)

and one East-West (horizontally) in the field of view. Use of a 40x objective is recommended.

Procedure

To insure the test is being done correctly, a slide of known monosodium urate crystals should be

used initially.

1. Swing the full wave plate out of the path of light.

2. Place the slide on the stage and bring crystals into a sharp focus. The needle shaped crystals

will appear white regardless of orientation.

3. Swing in the full wave plate and put the orientation handle in it’s extreme left position. Crystals

with a long dimension in the N/S direction should appear yellow and the E/W direction blue

(Figure 3).

4. Move the orientation handle to it’s extreme right position. Now the N/S crystals should be

blue, and E/W yellow. (Figure 3).

Be sure to test crystals with the orientation handle in each position to insure positive

identification.

ENGLISH

The following is the procedure for identification of pseudo gout:

The test for pseudo gout is done identically to the test for gout. However, the color change is

opposite that of Gout. That is, with the handle at the left extreme, N/S crystals are blue and E/W

crystals are yellow, and vice versa with the level at the right side (Figure 4).

1.0 Einleitung
